Ok, maybe later I will figure out why the chRNA effects sizes are positive in both productive and non-productive group… Porbably has something to do with library normalization.
Next I want to make some figures related to the splicing druggable genome. This paper has a nice supplemental table of small molecule “druggable” proteins that I can use to compare… I expect the splicing-modified genes to be not particularly enriched in any caetgory, and be more or less totally independent with the list of “druggable” proteins… The only hypothesis I do have (which I will test below) about what genes are more druggable by splicing-mediated expression effects are that longer genes (with more introns/exons, more potential for cryptic sites) will be enriched.
I think to do this, it will be uesful to make a list of splicing-mediated-risdiplam-effected genes, then an expression-matched list of control genes.

Next want to check enrichment of Tier1 (drugged genes), SplicingPerturbed, SplicingPerturbed control in different gene categories (ie kinase, nuclear receptors,GPCRs etc). I will define the categories based on GO categories, as retrieved from msigdb. I expected the Tier1 genes to be enriched in (non-olfactory) GPCRs, nuclear hormone receptors, kinases, and ion channels, while I expect Splicing perturbed to show no such enrichment.
